Development of a real-time duplex PCR assay for simultaneously diagnosis of KHV and CEV in common carp and koi

Koi herpesvirus (KHV) and carp edema virus (CEV) are potential risks for the koi trade global common aquaculture. Due to severe impact of both viruses, a robust diagnostic tool was required, capable detecting distinguishing infections with high accuracy precision. The aim this study describe real-time duplex TaqMan PCR assay simultaneous detection KHV CEV in koi. Two pairs primers two probes were used amplify specific conserved regions DNA. This confirmed be sensitive specific. Limit detections 15 DNA copies/µL 150 CEV, respectively. able identify distinguish KHV, but unable other pathogens sample matrices. For clinical validation, 18 positive samples each, as well 12 negative tested three different test methods, i.e., PCR, simplex conventional PCR. All tests provide optimal conformity results. results showed that detect presence each pathogen infected fish. is rapid, sensitive, fish, thus it can considered valid alternative aquaculture laboratories.

 ABSTRAK
 dan merupakan risiko potensial untuk perdagangan budidaya ikan mas global. Karena dampak dari keduanya yang parah, maka diperlukan perangkat diagnostik tangguh, mampu mendeteksi membedakan dengan akurasi presisi tinggi. Tujuan penelitian ini adalah menjabarkan pengujian real time dupleks pada secara simultan dalam satu reaksi. Dua pasang primer dua probe digunakan mengamplifikasi wilayah spesifik lestari CEV. Pengujian terbukti sensitif spesifik. Sensitivitas kopi DNA/µL mengidentifikasi tetapi tidak dapat patogen lain matriks sampel. Untuk validasi klinis, masing-masing sampel positif serta negatif diuji tiga metode berbeda, yaitu dupleks, tunggal konvensional. Ketiga tersebut memberikan kesesuaian hasil optimal. Hasil menunjukkan bahwa keberadaan setiap terinfeksi. Uji cepat, sensitif, mas, sehingga dipertimbangkan sebagai alternatif di laboratorium klinis akuakultur.
